TLDR; PROJECT SUMMARY AI BETA
This project uses computer simulations to study how potential drugs work against the main protein used by the COVID-19 virus to replicate. This helps scientists quickly find promising drug candidates for testing and development.
Note: This TLDR is a simplication and may not be 100% accurate.OFFICAL PROJECT DESCRIPTION
https://www.youtube.com/watch?v=VnyaAmM1nhE
These projects are to assist rapid sprints of relative alchemical free energy calculations for prioritizing compound designs from chemists from the COVID Moonshot for synthesis.
This series of projects simulates the SARS-CoV-2 main viral protease.
These projects collectively cover the entire set of X-ray structures collected by the COVID Moonshot, and more RUNs will be added as new structures are collected.
To learn more about what the Moonshot is and how it came about, you can read our blog post or watch this video.
In addition to helping us prioritize compounds, you can help us purchase more compounds for synthesis at cost from Enamine by sponsoring our GoFundMe page for patent-free open science COVID-19 drug discovery! This is a radical new approach to drug discovery that aims to rapidly produce inexpensive new therapies.

RELATED TERMS GLOSSARY AI BETA
alchemical free energy calculations
Calculations used to predict the binding affinity of molecules to a target protein.
Alchemical free energy calculations are a computational method used in drug discovery to estimate how well a molecule will bind to a specific protein. These calculations involve simulating the interactions between the molecule and the protein, considering factors like electrostatic forces, hydrogen bonding, and van der Waals interactions. The results provide insights into the binding affinity and can guide the design of new drugs with improved potency and selectivity.
compound designs
The process of creating new chemical structures with potential therapeutic properties.
Compound design is a crucial step in drug discovery where chemists create new molecules with specific biological activities. This involves synthesizing various compounds and evaluating their properties, such as their ability to bind to target proteins or inhibit specific enzymes. The goal is to identify promising candidates that can be further developed into potential drugs.
COVID Moonshot
A collaborative research initiative aimed at accelerating the development of effective treatments and vaccines for COVID-19.
The COVID Moonshot is a global effort to rapidly develop solutions for the COVID-19 pandemic. It involves scientists, researchers, and organizations from around the world working together to accelerate the discovery, development, and deployment of new therapies, diagnostics, and vaccines.
SARS-CoV-2
The virus responsible for the COVID-19 pandemic.
SARS-CoV-2 is a novel coronavirus that causes COVID-19. It is an RNA virus that primarily affects the respiratory system, leading to symptoms such as fever, cough, shortness of breath, and in severe cases, pneumonia and death.
main viral protease (Mpro)
A key enzyme produced by SARS-CoV-2 that is essential for viral replication.
The main viral protease (Mpro), also known as 3CLpro, is a critical enzyme involved in the replication of the SARS-CoV-2 virus. It cleaves viral proteins into smaller functional units, enabling the assembly and release of new virus particles. Inhibition of Mpro is therefore a promising target for antiviral therapies.
X-ray structures
3-dimensional representations of molecules obtained using X-ray crystallography.
X-ray structures are detailed 3D models of molecules determined through X-ray crystallography. This technique involves exposing crystals of the molecule to X-rays and analyzing the resulting diffraction patterns. The resulting data can be used to determine the precise arrangement of atoms within the molecule, providing valuable insights into its structure and function.
Enamine
A company that provides a wide range of chemical compounds for research and development.
Enamine is a leading supplier of chemical building blocks and reagents used in pharmaceutical and biotechnology research. They offer a vast catalog of commercially available compounds, as well as custom synthesis services to meet specific research needs.
GoFundMe
A crowdfunding platform that allows individuals and organizations to raise money for various causes.
GoFundMe is a popular online platform where individuals and organizations can create fundraising campaigns to collect donations from the public. It is used for a wide range of purposes, including personal emergencies, medical expenses, community projects, and charitable initiatives.
